SRX3510908: RNA-Seq of Chloridea virescens: adult female antennae
1 ILLUMINA (Illumina HiSeq 2500) run: 47M spots, 11.8G bases, 6.3Gb downloads

Design: Adult female Chloridea virescens were sacrificed by freezing at -80C. Antennae were subsequently removed from a single female just prior to RNA isolation. 2 antennae (one pair) were used for extraction. RNA was isolated according to Trizol protocol, then processed on RNEasy micro columns for clean up. RNA was quantified using Bioanalyzer and Qubit, the sent to the NY Genome Center for library preparation and sequencing. Libraries were prepared using the Ovation RNA-Seq System for preparing amplified cDNA from total RNA followed by Illumina TruSeq Nano DNA Sample Preparation. 125bp paired-end sequencing was performed on an Illumina HiSeq 2500.

We examined the transcriptomes of antennal and thoracic tissue from two closely related Chloridea (Lepidoptera:Noctuidae:Heliothinae) species. Chloridea virescens is a broad generalist pest of economic importance, while Chloridea subflexa is a narrow specialist on plants in the genus Physalis. To identify genes that are expressed in male and/or female antennae from one or both species but not in non-sensory tissues, we sequenced male and female antennae from each species, and from combined male+female thoracic tissue from each species.
